It depends on your body type actually. If you are very skinny and naturally dont put on weight, then yes twice a day is fine, coupled with alot of healthy food and protein. That should give you around 3200+ calories. Definatley will help you put on weight.

If your just looking to bulk up, but your body can get fat if you eat the wrong foods. I would use it sparingly. As weight gainers typically have alot of sugar and fat.

I personally used a weight gainer and had great success using it twice a day. I was very skinny though. Judging by your screen name (tryn2grow) and the fact your taking a weight gainer im assuming your fairly small. So eating about 4 solid high protein meals a day + adding extra calories and protein with your gainer should work just fine for you.
