For maximum benefit I would dose twice per day, alternating days. I don't think it matters whether you dose before or after workouts. The effects are long-term which is why you have to take it for long periods to see the benefits; but the effects are truly remarkable. (Naturaly hGH is released at night while you sleep, so you might want to take the second dose (pulse) right before bed to mimic the natural release pattern.)

Just as CJP says there really aren't many negatives. I guess if you really took way too much for way too long, you might eventually see growth abnormalities/deformities.
